Engineered with alternating aromatic and cationic residues to exploit the electrochemical gradient across the inner mitochondrial membrane, SS-31 is a cell-permeable tetrapeptide that has become a primary pharmacological tool in mitochondrial cardiolipin research. Also known as Elamipretide, MTP-131, or Bendavia, carries the molecular formula C32H54N10O5 and a molecular weight of 675.85 g/mol. Its sequence D-Arg-2,6-Dmt-Lys-Phe-NH2 incorporates a D-arginine and a dimethyltyrosine residue. Produced via solid-phase peptide synthesis.
In vitro studies using isolated mitochondria and submitochondrial particles have examined whether MTP-131 stabilises cardiolipin-cytochrome c interactions and modulates electron transport chain complex activity. In vivo rodent models have investigated the mitochondrial uptake kinetics of Bendavia using fluorescently labelled analogues, mapping the relationship between membrane potential, SS-31 accumulation and respiratory control ratio.
