What is FOXO4-DRI (Proxofim)?

A synthetic cell-penetrating peptide engineered from a fragment of the FOXO4 protein, also known as Proxofim, FOXO4-DRI is studied as a senolytic that clears worn-out cells. Research focuses on how it breaks up the FOXO4-p53 interaction that lingering senescent cells depend on to survive.

Application of D-retro-inverso (DRI) peptidomimetic chemistry to a FOXO4-derived interaction domain produced a 46-amino acid protease-resistant construct that has become a primary research tool in cellular senescence biology. FOXO4-DRI, also designated Proxofim, has an approximate molecular weight of 5400 g/mol. Produced via solid-phase peptide synthesis.

In vitro co-immunoprecipitation and fluorescence polarisation assays have investigated the binding affinity of Proxofim for the p53 interaction domain, examining whether the DRI construct competitively displaces endogenous FOXO4 from the FOXO4-p53 complex in senescent fibroblast cultures. In vivo rodent models have explored the pharmacokinetics and tissue distribution of Proxofim, examining whether the protease-resistant DRI backbone achieves sufficient systemic exposure and tissue penetration.
