Within the TGF-beta superfamily, few ligands have attracted as much preclinical attention for their role in negative regulatory signalling as Growth Differentiation Factor 8. GDF-8, more widely known as Myostatin, is a recombinant homodimeric protein with an approximate molecular weight of 25,000 g/mol. Myostatin signals through the activin type IIB receptor (ActRIIB) and downstream SMAD2/3 transcription factors. Produced as a recombinant protein.

In vitro reporter gene assays using SMAD-responsive luciferase constructs have been employed to characterise dose-dependent transcriptional activation initiated by recombinant GDF-8. In vivo rodent models carrying targeted Myostatin gene disruptions have provided comparative data for studies administering exogenous GDF-8.
