What is PTD-DBM?

Chimeric fusion of a cell-penetrating TAT protein transduction domain with a Dishevelled-binding motif produced PTD-DBM, a bifunctional construct engineered to deliver an intracellular Wnt pathway interaction domain across cell membranes without requiring transfection. Also known as TAT-DBM chimeric peptide, approximate molecular weight of 2000 g/mol. Produced via solid-phase peptide synthesis.

In vitro studies using Wnt-responsive reporter cell lines have investigated whether PTD-DBM competitively inhibits Dvl-Frizzled interaction and reduces TCF/LEF-dependent transcriptional output in a dose-dependent manner. In vivo rodent models have explored the biodistribution and cellular uptake efficiency of PTD-DBM in tissues with active Wnt signalling, examining whether the TAT-mediated delivery strategy achieves sufficient intracellular concentrations to modulate Dvl-dependent beta-catenin stabilisation.
