What is PE-22-28?

Derived from a minimal active fragment of the sortilin propeptide spadin, the seven-amino acid peptide PE-22-28 was designed as a selective inhibitor of the TREK-1 two-pore domain potassium channel. Also known as Mini-Spadin, approximate molecular weight of 800 g/mol. Produced via solid-phase peptide synthesis.
In vitro patch-clamp electrophysiology studies using TREK-1-expressing HEK293 cells have characterised the inhibitory potency and selectivity of PE-22-28 across the two-pore domain potassium channel family, examining whether the compound discriminates between TREK-1, TREK-2 and TRAAK subtypes. In vivo rodent behavioural models have investigated whether central administration of PE-22-28 modulates hippocampal neuronal excitability and synaptic plasticity parameters.
