What is P21 (P021)?
A synthetic peptide derived from the most active region of the nerve-growth factor CNTF, also known as P021, P21 is studied for its support of new brain-cell formation. Rational design based on the ciliary neurotrophic factor (CNTF) C-terminal domain produced P21, a small peptide mimetic that has attracted preclinical interest for its capacity to engage neurotrophic signalling pathways without requiring the full-length cytokine. Also designated P021 or CNTF peptide mimetic, approximate molecular weight of 500 g/mol. Produced via solid-phase peptide synthesis.
In vitro studies using primary hippocampal neuron cultures have examined whether P21 activates JAK/STAT3 and PI3K/Akt pathways downstream of the tripartite CNTF receptor. In vivo rodent models have investigated the oral bioavailability and CNS penetration of P21, examining whether this low-molecular-weight mimetic achieves central nervous system exposure sufficient to modulate dentate gyrus neurogenesis markers.
