What is Orexin B?
A synthetic 28-amino-acid brain peptide also known as hypocretin-2, Orexin B is studied as a receptor-selective counterpart to Orexin A. Research focuses on its preference for one of the two orexin receptors, making it a tool for separating their distinct roles.

Receptor subtype selectivity within the orexinergic system is most clearly investigated using the 28-amino acid neuropeptide Orexin B, which exhibits preferential affinity for the OX2R subtype over OX1R. Also designated Hypocretin-2 (CAS 205640-91-1), this peptide carries the molecular formula C130H211N39O33S2 and a molecular weight of 2937.42 g/mol. Unlike Orexin A, Hypocretin-2 lacks intramolecular disulphide bonds and the N-terminal pyroglutamate cap. Produced via solid-phase peptide synthesis.
In vitro comparative studies using cell lines expressing OX1R or OX2R in isolation have measured the differential potency of Hypocretin-2 versus Orexin A at each subtype, examining whether OX2R-selective activation produces qualitatively distinct downstream signalling patterns. In vivo rodent models have investigated whether selective OX2R engagement by Orexin B produces distinct patterns of neuronal activation in tuberomammillary and locus coeruleus nuclei.
