Encoded within the mitochondrial 16S rRNA gene, Humanin is a 21-amino acid mitochondrial-derived peptide that challenged the prevailing assumption that functional open reading frames within mitochondrial rRNA genes were non-existent. Also designated HN or MTRNR2, approximate molecular weight of 2690 g/mol and the sequence MAPRGFSCLLLLTSEIDLPVKRRA. Produced via solid-phase peptide synthesis.
In vitro studies using neuronal cell lines exposed to apoptotic stimuli have investigated whether Humanin modulates Bax translocation to mitochondrial membranes and subsequent cytochrome c release through direct protein-protein interaction. In vivo rodent models have explored the receptor-mediated signalling axis of HN, examining its reported interaction with the CNTFR/WSX-1/gp130 tripartite receptor complex and downstream STAT3 phosphorylation.
