Encoded within the mitochondrial genome, MOTS-c represents a rare class of mitochondrial-derived peptide that has opened a novel research frontier in retrograde mitochondrial signalling. Also known as Mitochondrial Open Reading Frame of 12S rRNA-c, this 16-amino acid peptide has the sequence MRWQEMGYIFYPRKLR and an approximate molecular weight of 1817 g/mol. Produced via solid-phase peptide synthesis.
In vitro studies have investigated whether Mitochondrial ORF 12S rRNA-c depletes intracellular 5-methyltetrahydrofolate and drives endogenous AICAR accumulation, thereby activating AMPK through an indirect metabolite-mediated mechanism. In vivo rodent exercise physiology models have explored whether exogenous MOTS-c administration alters skeletal muscle AMPK phosphorylation status and downstream ACC/mTORC1 signalling.
