What is ADAMAX?
A synthetic nine-amino-acid designer analogue of the peptide Semax, ADAMAX is studied for its effects on memory, learning and the survival of nerve cells. Research focuses on two structural additions, an acetyl cap and a rigid adamantane cage, that increase stability and the ability to cross membranes.

Building upon the ACTH(4-10) melanocortin fragment scaffold, the addition of both an N-terminal acetyl group and a C-terminal adamantylglycine residue produced ADAMAX, a structurally reinforced analogue designed for maximal enzymatic stability. Also known as N-Acetyl Semax Adamantylglycine, approximate molecular weight of 1000 g/mol. Produced via solid-phase peptide synthesis.
In vitro comparative degradation assays using serum and brain homogenate preparations have measured the proteolytic half-life of ADAMAX alongside ACTH(4-10)-Pro-Gly-Pro and unmodified ACTH(4-10), establishing a structure-stability hierarchy. In vivo rodent models have investigated whether the adamantylglycine modification alters CNS penetration or tissue distribution patterns compared to the parent compound.
