What is B7-33?
A synthetic single-chain peptide derived from the hormone relaxin-2, B7-33 is studied for its effects on tissue scarring and cardiovascular function. Research focuses on its biased signalling, weakly activating one pathway while strongly driving another and increasing a tissue-remodelling enzyme.

Reduction of the two-chain disulphide-bonded relaxin-2 structure to a single 26-amino acid chain produced B7-33, a minimalist agonist that has become a central tool for investigating biased signalling at the relaxin family peptide receptor 1 (RXFP1). Also designated RXFP1 biased agonist, approximate molecular weight of 2800 g/mol. Produced via solid-phase peptide synthesis.
In vitro studies using RXFP1-expressing HEK293 cells have compared the signalling fingerprint of B7-33 against native relaxin-2, examining differential activation of cAMP accumulation, ERK1/2 phosphorylation and beta-arrestin recruitment. In vivo rodent fibrosis models have investigated whether the biased signalling profile of B7-33 produces a pathway activation pattern distinct from that of full-length relaxin-2.
