B7-33 is a synthetic 26 amino acid single-chain peptide derived from the B-chain of human relaxin-2, engineered to act as a selective biased agonist at the relaxin family peptide receptor 1 (RXFP1). Unlike native relaxin-2, B7-33 activates RXFP1-mediated cAMP-independent signalling pathways — specifically pERK1/2 and pAKT — while minimising cAMP accumulation, making it a valuable pharmacological tool for dissecting RXFP1 signalling bias in vitro.
