To facilitate high throughput screening of TACE inhibitors, AnaSpec synthesized a novel peptide substrate for TACE using QXL™ 520/5-FAM FRET pair. Using this FRET substrate, AnaSpec developed a new kit – the SensoLyte™ 520 TACE Activity Assay. This kit can be used to detect the activity of the enzyme and for screening of TACE inhibitors. It is highly sensitive and can detect subnanogram amounts of enzyme.

To facilitate high throughput screening of renin inhibitors, we have developed new renin assay kits utilizing FRET peptide substrates. The SensoLyte™ 520 Renin Assay Kit contains FRET peptide labeled with the quencher QXL™520 and the fluorophore 5-FAM. These newly developed assays were validated with several known renin inhibitors. Assay with QXL™ 520/5-FAM substrate was approximately 40 fold more sensitive than an existing assay which uses EDANS/DABCYL FRET substrate.
Facile Synthesis of Hydrocarbon-Stapled Peptides

This poster describes a versatile synthesis method for hydrocarbon-stapled peptides. Asymmetric synthesis of (S)-Fmoc-a-(2’-pentenyl)-alanine was successfully accomplished via an Ala-Ni (II)-BPB-complex [3] in three steps with a 40% total yield. The 12-mer peptide containing two a- pentenyl-alanines on positions 4 and 8 was synthesized by Fmoc solid phase synthesis method. After olefin metathesis and cleavage, the peptide was purified by HPLC to obtain the hydrocarbon-stapled peptide.

In order to facilitate high throughput screening of AD drug candidates, we have developed a new SensoLyte™ 520 b-secretase assay kit using a fluorescence resonance energy transfer (FRET) peptide, HiLyte Fluor™ 488-Glu-Val-Asn-Leu-Asp-Ala-Glu-Phe-Lys(QXL™ 520)-OH. The longer excitation and emission wavelengths of HiLyte Fluor™488 minimize the interference from autofluorescence and absorbance of test compounds.
Solid Phase Synthesis of a Fluorescent Peptide: Comparison of Fmoc-Lys(5-FAM)-Resin and Fmoc-Lys[5-FAM(Trt)-Resin

We have prepared and performed comparison studies using two kinds of resins, Fmoc-Lys(5-FAM)-Rink Amide resin (I) and Fmoc-Lys[5-FAM(trt)]-Rink Amide resin (II), the latter contains a phenolic hydroxyl group protected with a trityl group. Syntheses were carried out under the same standard conditions and the peptides obtained showed no significant difference in purity. The results of these studies showed that resin (I) is adequate for synthesis of C-terminal fluorescent labeled peptide.
